A new 30-color spectral flow cytometry panel for T cell characterization. Draper D et al. AACR2025

The development of a 30-color immunophenotyping panel using the Cytek Aurora™ Spectral Flow Cytometer has enabled detailed profiling of T cell, NK and B cell subsets, along with their functional states. This panel includes markers for activation, exhaustion, cytotoxicity and proliferation, validated through comparisons between stimulated and unstimulated PBMCs. The study demonstrates memory T cell phenotyping and highlights significant changes in activation and exhaustion markers including PD-1, LAG-3, CD25, ICOS and Granzyme B to demonstrate the utility of spectral flow cytometry in assessing T cell therapies in both preclinical and clinical settings.
